What is Antibacterial Powder?
This powder is an antibiotic frequently utilized to manage bacterial infections. In Agar mycelium cultures, it effectively inhibits bacteria to a manageable level. It is employed to purify, germinate, isolate, and manipulate mushroom mycelium, significantly reducing contamination risks due to its antibacterial properties. A notable feature is its autoclave resistance, which simplifies usage for cultivators.
How Does This Powder Work?
Often, spores come with attached bacteria that transfer to the Agar media, resulting in fast-growing bacterial spores that hinder the germination of slower-growing mushroom spores. When these spores are introduced to agar media containing this antibacterial powder, the mushroom spores can thrive and develop, leaving bacterial contamination behind. The hyphae cells have long, sharp filaments that reduce bacterial carryover when passing through this antibacterial treatment.
Main Uses of Antibacterial Powder Through Agar Plates:
- Purify contaminated Agar plates through Agar transfer.
- Autoclave resistant; added to Agar mix prior to inoculation.
- Germinate spores from a spore syringe or spore print likely contaminated.
- Clone wild mushroom tissue prone to contamination.
Instructions for Making Agar Mix with Antibacterial Powder in Petri Dishes:
- Add 0.05g (1/20th of a gram) of antibacterial powder to your Agar mix for every 42g of Agar Mix (MEA/MYA/PDA, etc.).
- Boil 1000ml of hot water (for 42 gm of Agar mix) and mix in your antibacterial Agar.
- Stir thoroughly with a spoon. Pour slowly to avoid air bubbles.
- Autoclave or pressure cook for 20 minutes at 15 psi.
- Pour into Petri dishes at approximately 50 degrees Celsius and let set for 10 minutes. Avoid stirring post-autoclave to prevent air bubbles.
- Once Petri dishes are set, inoculate and wrap with parafilm, preferably after 3 days to ensure no contamination.
- Store Petri dishes upside down to minimize condensation.
